Mitochondria are the source of energy in cells, and their morphology can vary greatly (from rod-shaped to round). Mitochondria are very small and can only be observed by electron microscope. Mitochondria contain inherited maternal DNA. The number, size and shape of these cells vary according to animal cell types. Electron microscopy is the best way to observe mitochondria. Histopathological methods such as Altmann technique are helpful. The successful display of mitochondria by histochemical method depends on the following factors: the tissue must be fresh and fixed, and the slice is thin (2-3μm).
Aniline Acid Fuchsin Saturated Solution is the main staining reagent of Altmann method.
