Sephadex G-type glucan gel is only suitable for use in the aqueous phase, and the hydroxylated Sephadex LH-20 is suitable for separation of lipophilic molecules in organic solvents and purification of natural products in organic solvents.
The separation of LH-20 from glucan gel is mainly by gel filtration and has the function of reverse phase distribution.
Under the action of gel filtration, the retention of macromolecular compounds is weak, and they are eluted first, and the smallest molecules exit the column last. In the reverse-phase eluting solvent, the reverse-phase distribution effect is played. The more polar substances are eluted first, and the less polar compounds are eluted later.
Application:
The use of molecular sieve is suitable for the separation of lipophilic molecules with organic solvents, and various natural products can be prepared economically on a large scale. Such as: steroids, terpenes, lipids and small molecule polypeptides

Instructions for Use
1. Preparation of gel suspension
Soak the dry powder in 60-70% ethanol, stir well overnight, wash away any residue that may be present, and balance at least two column volumes according to your own eluent until the baseline is stable.
2. Eluting solvent
If the polarity of the sample is large, use reverse-phase solvent elution (methanol-water), dissolve the sample with the least volume of methanol-water, and filter the upper column; If the sample polarity is low, use chloroform-methanol positive phase elution.
3. Sample handling
Dissolve the sample with as little eluting reagent as possible and filter under atmospheric pressure.
4. Wet loading

Control the flow rate, generally less than 1drop/s.
