Sometimes the appropriate concentration of antibiotics is added to the cell medium, which can effectively prevent microbial contamination. At present, the most commonly used antibiotic is penicillin-streptomycin mixed solution (100× double antibody). Penicillin-Streptomycin-Gentamicin Solution (100 x) is a triantibody specifically designed for cell culture and can be added directly to cell culture after filtration. The content of penicillin, streptomycin and gentamicin in the mixed solution of penicillin-streptomycin (100× three antibodies) was 10kU/ml, the content of streptomycin was 10mg/ml and the content of gentamicin was 5mg/ml. The solution was prepared with 0.9%NaCl or PBS, and the recommended working concentration of penicillins in cell culture medium was 100U/ml, the working concentration of streptomycin was 0.1mg/ml, and the working concentration of gentamicin was 50μg/ml, that is, it could be used in a 100-fold dilution. A package of 100ml penicillin-streptomycin - gentamicin mixture (100× triple antibody) can prepare 10L cell culture solution.
Penicillin-streptomycin - gentamicin mixed solution (100× triple antibody) can be used in one of the following two ways:
1. Directly add in sterile cell culture medium: add penicillin-streptomycin and gentamicin mixed solution (100× triantibody) at the ratio of 5ml per 500ml cell culture medium, and mix well for use.
2. When preparing cell culture medium, add it, and then filter and remove bacteria: when preparing cell culture medium, add penicillin-streptomycin - gentamicin mixed solution (100× trianti) in the ratio of 10ml for each 1L cell culture medium, and filter and remove bacteria after preparation.
