Description:
Immunosuppressant. Cyclosporin A, also known as Cyclosporine or Antibiotic S 7481F1, is a commonly used immunosuppressive drug used to stop rejection in organ and bone marrow transplants. Cyclosporin A reduces immune system activity by interfering with T cell activity and growth [1][2]. Cyclosporin A binds to the intracellular receptor cyclophilin of lymphocytes, especially T cells, to form a complex surface that specifically targets calcineurin (phosphatase 2B) activity with an IC50 of 5 nM. Cyclosporin A also inhibits IL-2 production in activated T cells, with an IC50 of 10 nM[3]. Whether cultured FJ17 cells in vitro or FJ17-transfected mouse models, cyclosporin A can inhibit the production of interleukin IL-2 and IL-5, and results show that cyclosporin A can inhibit airway eosinophilic allergic inflammation by inhibiting the synthesis of IL-5 in T cells [4]. cyclosporin A affects mitochondrial function by interfering with the opening of mitochondrial permeability transport pore (MPTP), thereby inhibiting the release of cytochrome c and regulating apoptosis [5].

Application example (for reference only) :
1) To detect the inhibitory activity of Cyclosporin A on calcineurin, Jurkat cells were treated with 100 nM Cyclosporin A for 1h, and then the phosphatase activity was determined. It was found that Cyclosporin A effectively inhibited the Ca2+ dependent phosphatase activity. In addition, Jurkat cells were incubated in the medium containing OKT3 and PMA and different concentrations of Cyclosporin A (0.001-10000nM) for 24h to detect the activity of IL-2 products, and it was found that the IC50 value of Cyclosporin A to inhibit IL-2 was 10nM[3].
2) To test the effect of Cyclosporin A on cytokines in T cells in vitro, FJ17 was cultured with mitomycin-C treated spleen cells for 24h, and Cyclsporin A (1,10,100nM) of different concentrations were added to the culture system. IL-2, IL-4, and IL-5 products were detected and Cyclosporin A was found to inhibit IL-2, IL-4, and IL-5 products in a dose-dependent manner. In addition, 30mg/kg Cyclosporin A was found to significantly inhibit the mRNA expression of IL-2 and IL-5 in vivo [4].

This product is highly soluble in methanol, ethanol or acetonitrile, easily soluble in ethyl acetate, soluble in acetone or ether, and almost insoluble in water.
