Principle:

Polylysine solution is a widely used adhesive for tissue sections and slides. The interaction of polycationic molecules with anions on tissue sections can produce strong adhesion. The nature of the culture bottle surface is very important for cell culture, the glycoprotein (negative) on the cell surface is easy to adsorb on the hydrophilic surface, so if there is a considerable amount of positive charge on the cell culture surface, it can promote cell adsorption, which is the important place to optimize the cell culture surface with polylysine.

Application:

It is suitable for anti-slip treatment of slides used in histology, immunohistochemistry, frozen section, cell smear, in situ hybridization, etc., to prevent tissue from falling out during experimental operation. It can also be used in cell culture to increase cell adhesion ability.

Slide processing:

1. Dilute polylysine solution with sterilized water, and the general use concentration is 0.01%.

2. Soak the slide in diluted polylysine solution for 5 minutes. Note that adding time does not improve the coating effect.

3. Dry in 60℃ oven for 1 hour, or overnight dry at room temperature 18-26℃.

Note:

1. Every 100mL of diluted polylysine solution should be covered by 40-90 slides, more than 90 slides will affect its adhesion.

2. The slides must be kept clean before use. If necessary, wash with 70% ethanol solution containing 1% HCl.

3. The released polylysine solution should be placed at 2-8 ℃ and is stable for at least 3 months.

4. The used diluent should be filtered and discarded if there is turbidity or bacteria

5. The concentration of polylysine can be higher, and the sucked out can be reused at least 20 times.

Culture bottle treatment:

1. Coated with culture plate or culture bottle, the polylysine concentration is generally 0.01%,

2. Prepare 100ml of steam water and autoclave it.

3. 0.01% of polylysine was evenly applied to the surface of the culture substrate at an amount of 50 microliters per square centimeter, and left for 5 minutes at room temperature to absorb excess liquid.

4. Add sterilized water, rinse repeatedly three times, aseptic operation.

5. Sterile and dry after treatment.

6. Polylysine that remains sterile after recovery can be reused repeatedly.

Commonly used compound substances can be divided into five categories:

1, alcohol, the use of 30 ~ 70%, mainly used in various egg products.

2, organic acids, often used organic acids are generally: acetic acid, malic acid, maleic acid, citric acid, succinic acid, etc., the use of between 0.5 and 50%, mainly used in rice, drinks, salads, sauces and other foods;

3, triglyceride, triglyceride is mostly low fatty acid ester, the amount is between 0.01 and 5%, mainly used in animal protein, milk protein more food.

4, glycine, the dosage is 0.01 ~ 10%; Mainly used in milk preservative.

5, other natural antibacterial agents, such as: protamine, tea polyphenols, etc.
