Product Introduction:
NBT is Nitroblue tetrazolium chloride, a dark blue amorphous slightly soluble substance. BCIP, also known as 5-Bromo-4-Chloro-3-Indolyl Phosphate, is catalyzed by Alkaline phosphatase (AP).
BCIP is hydrolyzed to produce a highly reactive product that reacts with NBT to form an insoluble dark blue to bluish-purple precipitate.
Instructions for use:
1. Take 40ul 25×NBT and add it to 1ml 1×AP reaction buffer, mix it well, and then add 40ul 25×BCIP to mix it well, that is, to form a reaction working liquid. This reaction liquid is best used now. Note: Depending on the membrane size, the amounts of 1×AP reaction buffer, 25×BCIP, and 25×NBT can be scaled up equally.
2. Dip the washed PVDF/NC film into the reaction working liquid and gently shake it at room temperature for warming.
3. When the color depth of the protein band meets the requirements (generally about 20-30 minutes), the membrane is immersed in deionized water to stop the reaction.
Note:
BCIP and NBT are suspected carcinogens, please take necessary preventive measures. Wear gloves during operation, avoid contact with skin as much as possible, and rinse thoroughly after use.
