Explanation: Ethylenediaminetetraacetic acid disodium is a divalent cation chelator, which can inhibit the activity of enzymes, such as metalloproteinases, because metalloproteinases require the presence of divalent cations to be active.
Preparation of 0.5M EDTA (pH=8.0) :
0.5M EDTA-Na2:9.306g dissolved in 50mL water, initial PH of stock =4.77, initial PH of sample =4.44, PH was adjusted to 8.0, and completely dissolved into colorless clear solution. Use after autoclaving.
Notes:
(1) EDTA-Na2.2H2O is not easily soluble in water and can only be dissolved under alkaline conditions. When the pH is adjusted to 8, it can be completely dissolved.
(2) Pay attention to adjust the pH of the final volume solution to 8.0.
