Multiple Western blots of the same membrane can be performed using the membrane regeneration solution. At room temperature, the used membrane is soaked in the membrane regeneration solution for 10-60 minutes, which can remove the bound antibodies on the membrane without affecting the antigen proteins. The next round of Western Blot experiments can then be performed using different antibodies. Suitable for multiple detection of different proteins from a small number of samples. Even if the sample size is not scarce, using this strategy to detect proteins on the same membrane multiple times can eliminate costly steps such as drug administration, protein electrophoresis and membrane transfer. Moreover, the error caused by re-loading can be eliminated, so that the comparability is stronger.

Western Blot film regeneration solution features:

Non-toxic and harmless, use at room temperature, store at room temperature; More than 5 Western Blot tests can be performed on the same membrane.

Application: Clear antibodies bound to nitrocellulose membranes or PVDF membranes without affecting antigen proteins.

Usage:

1. The film is fully immersed in the appropriate volume of regenerated liquid, room temperature elution 10-20 minutes, the length of time should be optimized by referring to the precautions, elution of some antibodies takes a longer time, more up to 60 minutes, if the room temperature elution effect is not good, the film regenerated liquid can be heated to 50 degrees, elution under 50 degrees water bath conditions.

2. Remove the membrane and rinse twice with washing liquid (such as TBST, etc.).

3. At this time, the antibody on the membrane has been removed, and another antibody can be added for detection after re-sealing.

Notes: 1. The essence of the membrane regeneration solution is to elute the antibody without affecting the antigen bound on the membrane. In actual operation, the membrane type, antibody type, concentration and antigen characteristics all affect the elution difficulty.

2. Suitable for Western detection of ECL and similar chemiluminescent reagents. Not applicable to Western tests performed with non-chemiluminescent reagents, such as DAB, NBT/BCIP.

3. After ECL color development, after elution of the regenerated solution for a few minutes, the ECL color development solution can be added again. If the strip is displayed on the film, indicating that the antibody is not cleaned, it must be further soaked in the regenerated solution until there is no color development on the film.

4. The membrane closed with skim milk powder is easier to regenerate than the membrane closed with BSA.

5. Try to avoid drying the membrane.
