Bacterial protein extraction kit is used to extract soluble protein from Escherichia coli. The lysate in the kit contains protease inhibitor, which has a strong effect and can quickly obtain total protein. It can be used for basic research experiments such as western blot experiments. Bacterial protein extraction kit is only used for scientific research.

Operation method :

1. Extraction of bacterial soluble protein

10ul of protease inhibitor and PMSF were added into 1ml of cold lysate. Mix well and set aside on ice.

The bacteria were weighed by wet weight, and then the newly configured lysate was added according to the proportion (1g: 10ml), and mixed, paying attention to the operation on the ice during the process;

Transfer the mixed liquid into the ultrasonic instrument, ultrasound;

Ultrasonic conditions: total time 45min, ultrasonic time 3s, interval 3s, process in ice bath environment;

After ultrasound, w4 degree centrifugation, 10000r/min, 20min;

Absorb the supernatant into the new tube;

Carry out protein quantification or denaturation for protein experiments.

(The extracted protein is recommended to be stored at -80℃, ready to use, to avoid repeated freezing and thawing, to avoid long-term storage.)

Note:

During the experiment, all reagents need to be pre-cooled or melted to ensure a low temperature environment during the operation;

PSMF (toxic) is used now, because PMSF will degrade rapidly in an aqueous solution;
