Conclusion: Insulin is a protein hormone, which is secreted by pancreatic β cells in vivo. It is the only hormone in the body that lowers blood glucose, and it is the only hormone that simultaneously promotes the synthesis of glycogen, fat, and protein. It is mostly used in animal experiments.
Preparation and preservation of mother liquor:
It is insoluble in water at neutral pH. It can be dissolved in dilute acetic acid or dilute hydrochloric acid (pH 2-3) to prepare a mother liquor of 2mg/ml. The mother liquor was divided and frozen at -20℃ according to the single dose to avoid repeated freezing and thawing. In addition, the mother liquor removed by filtration with a low-protein adsorbent membrane, or with a suitable antibacterial agent such as 0.1% thiomersalate or sodium azide, can be stored at 2-8 ℃ for up to several months.
Notes:
1) Since insulin is non-sterile, it is recommended to filter the insulin solution with a low-protein adsorption filter membrane to remove bacteria, and autoclaving cannot be carried out.
2) This product is also soluble in 125mM NaHCO3. The preparation of alkaline storage solutions is not recommended because high pH values can enhance deamidation reactions and protein aggregation.
