Product Introduction:
Monensin, a natural ionophore, can selectively bind monovalent cations such as Li+, Na+, K+, Rb+, Ag+ and Tl+, and transport these cations to the cell membrane. Promote the death of Gram-negative bacteria (such as Micrococcus, Bacillus, and Staphylococcus) by affecting pH or Na+-K+ balance, slow down the proliferation of P. falciparum and Coccicdium protozoa, And inhibit the replication of some viruses. In addition, monensin can also destroy the Golgi apparatus and inhibit intracellular protein transport, so it is commonly used as a protein transport inhibitor to inhibit the secretion of secreted proteins such as cytokines, or to enhance the staining of secreted proteins by inhibiting secretion during immune staining.
Description:
Na+ ionophores; Blocking glycoprotein secretion; It may induce catecholamine secretion in chromaffin cells. It is used to study the potential and spectroscopy of alkali metal ion complexes.
Use and synthesis method:
Monenmycin is available as sodium salt in powder and solution. Monenmycin powder can be prepared as storage solution with organic solvent such as ethanol (25mg/ml). Monenamycin in solution form is 1000× concentrated solution dissolved in ethanol (concentration is 2mM). Monenmycin is slightly soluble in water, and its storage solution can be diluted in buffer or isotonic salt solution to enhance its water solubility. However, if subsequent biological experiments are carried out, it is necessary to ensure that the residual amount of organic reagents is reduced to a limit that does not affect the experiment, because organic solvents can affect physiological effects even at very low concentrations. It is recommended that monenmycin aqueous solution be prepared for immediate use and stored for no more than 1 day.
Monenamycin is commonly used in the concentration range of 1-5μM. The commonly used concentration of immunostaining for enhancing secreted proteins is 2μM, and pretreatment can be done for about 1 hour. The specific concentration depends on the experimental application.
Note :
(1) For cell experiments, after dissolution, disposable needle filter must be used to remove bacteria.
(2) For your safety and health, please wear a lab coat and wear disposable gloves.
