Product Introduction:
DiI also known as DiIC18 (3),The full name of the reagent is 1, 1 - dioctadecyl - 3 - tetramethylindocarbocyanine perchlorate, is one of the most commonly used the membrane of the fluorescent probe, orange red fluorescence. DiI is a lipophilic membrane dye. After entering the cell membrane, dii can diffuse laterally and gradually stain the whole cell membrane.DiI fluorescence is very weak before entering the cell membrane, and only after entering the cell membrane could be excited to fluorescence strongly. After DiI is excited, it can emit orange-red fluorescence. The excitation spectrum and emission spectrum of DiI and phosphoester bilayer are obtained. Among them, the maximum excitation wavelength is 549nm, and the maximum emission wavelength is 565nm.DiI can be dissolved in absolute ethanol, DMSO and DMF with a solubility of about 1-2.5mg/ml. If it is found to be difficult to dissolve, it can be heated appropriately and sonicated to promote dissolution.

Uses:
1. DiI is widely used as a tracer or long-termtracer for forward or reverse, living or fixed nerve cells or tissues. DiI usually did not affect cell viability. Dii-labeled neurons could survive for up to 4 weeks in vitro culture and up to a year in vivo. The migration rate of DiI on the membrane of fixed neurons is 0.2-0.6mm/day, and the migration rate of dii on the membrane of living neurons is 6mm/day. 2. DiI can also be used to detect cell fusion and adhesion, detect cell migration during development or transplantation, and detect the diffusion of lipid on the cell membrane by Fluorescence Recovery After Photobleaching (FRAP). Cytotoxicity and labeled lipoproteins are detected.

Precaution:
1. When is used for fluorescent labeling of cell membranes, the usual concentration of DiI is 1-25μM, and the most commonly used concentration is 5-10μM.
2. DiI can stain live cells or tissues directly, and the staining time is usually 5-20 minutes.
3. For fixed cells or tissues, it is generally appropriate to use 4% paraformaldehyde prepared in PBS for fixation, as the use of other inappropriate fixators may result in a high fluorescence background.
