When Thioflavin T binds to the beta lamellae, such as those in amyloid oligomers, the dye undergoes the characteristic 120nm redshift of its excitation spectrum, which can be selectively excited at 450nm to produce a 482nm fluorescence signal. Thioflavin T rapidly and specifically binds to antiparallel beta-folded fibril formed from synthetic beta-amyloid protein (1-40), but does not bind to monomers or oligomeric intermediates. Binding of Thioflavin T does not interfere with the aggregation of beta-amyloid peptides into amyloid fibril. The dye is often used to visualize plaques made up of beta-amyloid proteins found in the brains of people with Alzheimer's disease.
Examples of using this product(for reference only)
In Vitro:
Aβ25–35 aggregation assay(20 μM Aβ25–35;20 μM Th-T solution)
Th-T is a relatively common fluorescent dye for detecting Aβ aggregation. The experiment was divided into three parts.
①. Aβ25–35 aggregation assay: 20 μM of Aβ25–35 with and without baicalein (80 μM) were incubated at 37 ℃ for 6 days. After incubation, 10 μl of the mixture was added into 190 μl of Th-T solution (20 μM) in a black 96-well plate. The fluorescence intensity was measured daily at the same time for 6 consecutive days.
②. Anti-aggregation assay of baicalein: A mixture of Aβ25–35 (20 μM) and baicalein (40, 60 and 80μM) was prepared in PBS, then the mixture was incubated at 37 ℃ for 6 days. 10 μl of the mixture was added into 190 μl of Th-T solution (20 μM), and the fluorescence intensity of the mixture was then measured.
③. Disaggregation of Aβ25–35 amyloid fibrils by baicalein: Aβ25–35 (20 μM) solution was incubated alone for 3 days, then it was incubated with baicalein (40, 60 and 80 μM) for another 2 days at 37 ℃. After incubation 3 days or 5 days, 10 μl of the mixture was added into 190 μl of Th-T solution (20 μM), and the fluorescence intensity was measured using a microplate reader at 440 nm excitation and 490 nm emission.
