Preparation of reserve liquid
1mg of TMRM Perchlorate was dissolved in 399.257 uL DMSO to obtain 5 mM reserve solution.
Preparation of TMRM working fluid
The reserve solution was diluted with serum-free cell medium or PBS to obtain 1-20 μM working solution.
Note:
Please adjust the concentration of TMRM Perchlorate according to the actual situation.
Cell staining Suspended Cells (6-well plates)
a. Centrifuge 1000g at 4℃ for 3-5 minutes, discard the supernatant. Wash twice with PBS for 5 minutes each time. The cell density was 1×106/mL.
b. Add 1 mL of working liquid and incubate at room temperature for 5-30 minutes.
c. Centrifuge 400g at 4℃ for 3-4 minutes, discard the supernatant.
d. Wash twice with PBS for 5 minutes each time.
e. Serum-free cell medium or PBS were used for suspension cells. Fluorescence microscopy or flow cytometry were used.
Adherent cell
A. The adherent cells were cultured on a sterile cover slide.
b. Remove the cover glass from the medium and suck out excess medium.
c. Add 100μL working solution, shake gently to completely cover cells, and incubate at room temperature for 30-60 minutes.
d. Wash the medium twice for 5 minutes each time. Fluorescence microscopy or flow cytometry were used.
Note:
If flow cytometry is used, the cells need to be rehung before staining.
