Product Introduction
T5 exonuclease degrades DNA in the 5 '-3' direction, and it can degrade double-stranded DNA, single-stranded DNA and engraved plasmid DNA. It can degrade DNA both from the 5 '-terminal and from incisions or gaps in linear or circular double-stranded DNA, but cannot degrade superhelical double-stranded DNA. Based on the above characteristics, T5 exonuclease can be applied to Gibson assembly.
Precautions
(1) 1 x T5ExoBuffer: 50mMKAc, 20 mmtrIS-ACph7.9, 10 MMMG (Ac) 2,1 mMDTT. This enzyme is also active in the PCRBuffer.
(2) The optimal reaction temperature of the enzyme is 37℃, and it is also active at 50℃, so it can be assembled by Gibson.
