Determination of Significance:
Caspase is a family of proteases involved in the process of apoptosis, including more than 10 members. Caspase-2, also known as Ich-1 or Nedd-2, is activated in the process of apoptosis signal transduction. There are two different splicing variants of caspase-2 mRNA. The protein translated by the full-length mRNA can promote apoptosis, while the short protein product can inhibit apoptosis. Caspase 2 can be activated by caspase-1, caspase-3 and granzyme B.
Measurement Principle:
The caspase-2 colorimetric assay is based on the hydrolysis of the peptide substrate N-acetyl-Val-Asp-Val-Ala-Asp-pNA (Ac-VDVAD-pNA) by caspase-2, resulting in the release of the p-nitroaniline (pNA) moiety. p-Nitroaniline has a high absorbance at 405 nm. The activity of Caspase can be calculated by detecting pNA. This kit is suitable for mammalian tissue and cell.
