Biochemical reagent, biochemical experimental studies, Wortmannin is a potent and specific phosphatidylinositol 3-kinase (PI3-K) inhibitor with IC50 of 2-4 nM. It can inhibit the formation of autophagosomes and also effectively inhibit DNA-PK/ATM with IC50 of 16 nM and 150 nM, respectively.
Wortmannin can enhance the apoptotic effect induced by radiation or deserum culture and prevent the anti-apoptotic effect of cytokines by inhibiting the PI3-K/Akt signaling pathway. The inhibition of PI3-K by warmancin can also block many short-term metabolic effects caused by insulin receptor activation. Wortmannin has no obvious antibacterial activity, but has strong antifungal activity.
Wortmannin and LY294002 are commonly used inhibitors of PI3K; LY294002 is the first synthetic molecule to inhibit PI3Kα/δ/β with IC50 of 0.5 μM, 0.57 μM and 0.97 μM, respectively. It is more stable than Wortmannin in solution and also inhibits the formation of autophagosomes. In contrast with Wortmannin, LY294002 can cause changes in intracellular calcium ion concentration, while wortmannin usually does not, so it has different effects on other cell activities compared with wortmannin. If the study is closely related to intracellular calcium ion concentration, then the application of LY294002 should be more cautious (for reference only).
