This method avoids the conflict between the conditions of DNA denaturation and the simultaneous detection of intracellular and surface markers. Continuous BrdU incorporation can be used to identify and analyze cells in the active state of DNA synthesis from those in the quiescent state, and thus to analyze the proportion of cell proliferation; On the other hand, the addition of BrdU at different time points allowed the analysis of cell cycle kinetics.
