Urine containing free hemoglobin is called hemoglobinuria, which is transparent bright red (containing oxyhemoglobin) or dark red (containing methemoglobin). In severe cases, it is dark brown or soy sauce color, and the color does not change after centrifugation. There are no red blood cells in the sediment, and the occult blood test is positive. Free hemoglobin in the blood can increase in large amounts, when more than 1.00-1.35 g/L, hemoglobinuria can occur, this situation is common in blood group incompatibility transfusion, paroxysmal nocturnal hemoglobinuria, cold hemoglobinuria, acute hemolytic diseases, etc. It can also be seen in various viral infections, streptococcal septicemia, malaria, extensive burns, extracorporeal circulation, renal dialysis, and massive destruction of red blood cells caused by surgery. Therefore, the qualitative detection test of urine hemoglobin has a high clinical value.
This kit can also be called urine occult blood test. Its detection principle is that heme in hemoglobin catalyzes H2O2 as an electron acceptor to oxidize the chromogenic substrate (o-toluidine) into o-methylazobene, which is green or blue, and its color is proportional to the hemoglobin content. It is often used to test urine hemoglobin in human or animal urine.
