Cerebro-Spinal Fluid (CSF) is a colorless transparent fluid that exists in the cerebral ventricle, subarachnoid space, and central canal of the spinal cord. It is produced by the choroid plexus in the cerebral ventricle and has similar properties to plasma and lymph. The cerebrospinal fluid of a normal adult is about 100 to 150ml, weakly alkaline and devoid of red blood cells. Normal cerebrospinal fluid has a certain chemical composition and pressure, which plays an important role in maintaining the relative stability of intracranial pressure. When the central nervous system is damaged, the detection of cerebrospinal fluid becomes an important auxiliary diagnostic method. Total Protein (TP) is composed of albumin and globulin. The determination of TP in biological fluids (serum, urine, cerebrospinal fluid) is generally based on the following two assumptions: 1. All protein molecules are composed of pure peptides with a mass percentage of nitrogen content of 16%; 2. Body fluids contain hundreds of protein molecules, each of which has very similar characteristics to the assay reaction; At present, the commonly used methods are: biuret method, ultraviolet spectrophotometry, dye binding method, Kjeldahl nitrogen determination method, precipitation method, etc.
The detection principle of the kit is that under acidic conditions, first, pyrogaleol red and molybdic acid complexed to form a red complex, which forms a complex with protein, and its absorption peak is moved to 604nm. The protein content in the sample is obtained by the colorimetry method of the microplate detector, which is specially used for the determination of total protein content in human or animal cerebrospinal fluid samples. However, it is susceptible to surfactant.
