nucleic acid is a biological macromolecular compound composed of many nucleotides, which is one of the basic substances of life. It widely exists in all animal and plant cells and microorganisms. According to the chemical composition of different nucleic acids, it can be divided into DNA and RNA. The nucleic acid molecule contains a certain proportion of phosphorus, 9.2% in DNA and 9.0% in RNA. The detection principle of the kit is that the 2′ and 3′ carbon atoms of the 5′ -nucleotide ribogroup have hydroxyl groups, and the bond between these two carbon atoms is weak. Periodic acid can oxidize it to form a dialdehyde compound, which can form a compound with a methylamine group. The compound takes off the phosphate group under acidic conditions, and determines the amount of inorganic phosphorus, that is, 5′ -nucleotide. Inorganic phosphorus and ammonium molybdate form yellow ammonium phospomolybdate, and then the reducing agent reduces the high-valence molybdenum ion to low-valence molybdenum ion, thereby forming blue molybdenum blue. In a certain concentration range, the blue depth is proportional to the phosphorus content. The absorbance at 660nm is detected, and the phosphorus content is obtained by checking the standard curve. The inorganic phosphorus that has not been oxidized by iodate was determined and deducted to be the exact 5′ -nucleotide phosphorus. The kit is characterized by the specific determination of 5′ -nucleotides.
