Red blood cell enzyme defects can be detected by methemoglobin reduction test, ascorbic acid, Heinz bodies, etc. This kit uses Schumm method, and its detection principle is that in the presence of sufficient NADPH, methemoglobin can be reduced to ferrous hemoglobin by methemoglobin reductase. When the content of glucose-6-phosphate dehydrogenase in red cells was normal, the amount of NADPH produced by the pentose phosphate metabolic pathway was sufficient to complete the above reduction reaction, and when the G6PD content in red cells was insufficient or deficient, the reduction rate of methemoglobin was slowed down or even could not be reduced, and methemoglobin was brown, which was detected by spectrophotometer at 635nm.
