CFDA, SE is a kind of cell staining reagent that can fluorescently label living cells, which can be used not only for In Vitro
experiments of cell proliferation, but also for tracking cell division and proliferation process In Vivo. After penetrating the cell membrane into the cell, it is catalyzed by the intracellular esterase to decompose into CFSE, which can occasionally and irreversibly bind to the amino group in the cell to the cellular protein. During cell division and proliferation, CFSE labeled fluorescence can be equally distributed between the two daughter cells, so its fluorescence intensity is half that of the parent cells, and it can be analyzed at 490 nm excitation light by flow cytometry or fluorescence microscopy. CFDA, SE labeled cells can be observed In Vivo for several weeks. Therefore, CFDA and SE are often used for live cell assay and fluorescence electron microscopy to observe the long-term activity of cells.
Dyeing steps (for reference only) :
(1) Prepare 1mM CFDA, SE solution with DMSO. Dilute it with PBS or an appropriate buffer to a CFDA,SE solution of 10 ~ 50μM.
(2) CFDA, SE solution of 1/10 cell medium volume was added to the cell medium.
(3) Cultured cells at 37℃ for 15-30 minutes.
(4) Wash the cells twice with PBS or an appropriate buffer.
(5) The cells were observed by fluorescence microscopy with a filter with excitation wavelength of 490nm and emission wavelength of 530nm.
