Product Introduction:
Draq5 is a far-infrared fluorescent live-cell DNA dye that is an anthraquinone dye with high affinity for double-stranded DNA. It is a dye that can penetrate the cell membrane and can be used to label live or post-fixed/dead cells. In flow cytometry, this dye can be used to distinguish between nucleated and unnucleated cells. Because Draq5 binds to DNA in a stoichiometric manner, it can also be used to report nuclear DNA content, which is suitable for chromosome ploidy and cell cycle analysis. It can be used as a nuclear counterstain in fluorescence microscopy analysis. Draq5 has many applications and is highly compatible with widely used programs of existing instrument platforms. The main application areas of DRAQ5 are HCS, cell models, GFP, flow cytometry and fluorescence microscopy.
The excitation wavelength range of Draq5 is 488 to 647 nm. For imaging microscopy, a light source of 633 or 647 nm is recommended for excitation. For flow cytometry, when excited at 488 nm, this dye can be detected using a 685 LP dichloroscope and a 710/50 channel. When excited at 633 nm, the 660/20 channel can be used for detection. For cell cycle /DNA analysis applications, it is recommended to use filters with longer wavelengths, such as a 735 LP dichloroscope and a 780/60 channel to optimize CV values for G1 and G2/M peaks. Make sure your instrument can detect the dye. Due to its wide range of emission and excitation wavelengths, the combination of Draq5 with other far-red fluorescent dyes that can be excited by 488 or 633 nm lasers is not recommended. The product concentration is 5 mM, using 5×104 cells/well as an example, the staining working solution concentration is calculated at 20 uM, and the specification of 200 ul could do 400 samples.
Precaution:

2. Fluorescent dyes all have quenching problems, please try to avoid light to slow down fluorescence quenching.
