Cell Tracker CM-DiI is a derivative of DiI and is more soluble in water than DiI. It can freely cross the membrane into the cell and be converted into the membrane impermeable reaction product. Cell Tracker CM-DiI dye is retained in living cells through several generations of culture. Cell Tracker CM-DiI dye can emit fluorescence stably for at least 72 h, which has ideal tracking performance, and has advantages such as stable property, non-toxic working concentration, good cell retention, and bright fluorescence under physiological pH conditions. In addition, the excitation and emission spectra of Cell Tracker CM-DiI dyes are fully separated from GFP (Green fluorescent protein) spectra and can be reused in multiple colors.
Cell Tracker CM-DiI contains a mild sulfhydryl reactive chloromethyl substituent that can be cross-linked to an aldehyde-containing fixative by covalently binding to sulfhydryl peptides and proteins without loss of staining signal after fixation. Compared with DiI and other non-covalently bound dyes, it is more suitable for the staining of fixed cells.
Protocol (for reference only)
Step 1 Prepare
(1) Configure DMSO storage solution. (The unused storage solution is stored at -20℃ to avoid repeated freezing and thawing.)
(2) Working solution: Dilute the storage solution with a suitable buffer (such as serum-free medium, HBSS or PBS) and prepare a working solution with a concentration of 1 to 5 μM.
2. Cell staining
The cells were incubated at 37℃ with working solution for 60 min. After labeling, the cells were cleaned with PBS and re-suspended in fresh medium.
For adherent cells, labeling during adherent culture had higher cell viability than labeling after digestion into suspension.
The following steps are optional (for reference only) :
Cell Tracker CM-DiI staining is compatible with subsequent fixation and penetration.
(1) Fixed. Use 3.7% paraformaldehyde (dissolved in PBS) and fix at room temperature for 10 min. Wash twice with PBS at room temperature for 5 min each time.
(2) Penetration. Permeate with acetone at -20℃ for 10 min. Wash twice with PBS at room temperature for 5 min each time.
