Di-8-ANEPES belongs to the ANEP family of dyes and can be used to determine rapid membrane potential changes. ANEP dyes are a class of dyes that can quickly react to changes in membrane potential, and their optical reaction can quickly detect changes in transient transmembrane potential in cells, which can prove the transformation of membrane potential dependent excitation spectrum. This property can detect the change of membrane potential by changing the excitation rate. ANEP dyes have weak fluorescence in aqueous environments and can change to strong fluorescence in lipid environments (such as cell membranes). Fast response probes operate by changing the electronic structure to produce their fluorescence properties in response to changes in the surrounding electric field. Their optical response enables rapid detection of changes in millisecond transmembrane potential in excitable cells, such as single neurons, cardiomyocytes, and intact brains. Due to its sulfonated group, Di-8-ANEPPS is less affected by cell internalization than other ANEPP dyes and is more lipophilic, making it more suitable for long-term membrane potential studies. In addition, Di-8-ANEPPS is more photostable and less photosensitive than Di-4-ANEPPS.
