荧光素-5-马来酰亚胺是巯基检测试剂。在大多数蛋白质中，反应位点是半胱氨酸残基，其本质上存在或由胱氨酸的还原产生。与碘乙酰胺不同，马来酰亚胺在生理条件下不与组氨酸和甲硫氨酸反应。
