Cy7 (Cyanine 7) is a near-infrared anthocyanin fluorescent dye. Because its fluorescence wavelength (Em: 770nm) is in the region of the near infrared window I of body tissue, Cy7 is often used In Vivo imaging of small animals. Cy7 fluorescence is not visible to the naked eye and requires near-infrared filters for microscopic observation, which many fluorescence microscopes (including confocal microscopes) are not equipped with, but flow cytometry is often equipped with Cy7 filters. Cy7 is a commonly used fluorescent dye. Cy7 can be used to label nucleic acids, proteins, antibodies, peptides, etc. It is one of the common small animal developers.
NHS activated esters (N-hydroxysuccinimide ester, N-hydroxysuccinimide ester, succinimidyl ester) are commonly used as activated groups in biomarker reactions. It activates the carboxyl group in the Cy7 molecule, allowing it to react with the amine group (primary or secondary amine) on the target biomolecule to form a stable amide bond, thus labeling the Cy7 molecule to the biomolecule. Since free amines are common functional groups on the surfaces of proteins, antibodies, and polypeptides (derived from the lysamine side chain), Cy7-NHS can react directly with them. Nucleic acid molecules naturally do not carry free amines, and only RNA or DNA modified with amines can react with Cy7-NHS. In addition, Cy7-NHS can also label polymer amine groups, nanoparticle surface amine groups, etc.
* Note:
1) The NHS ester group is sensitive to humidity, so the product must be dried and preserved. The product should be brought to room temperature for at least 20 minutes before use.
2) Use no amino buffer system. Examples include PBS, Hepes, sodium bicarbonate buffers or boric acid buffers. Do not use any buffer containing primary or secondary amines, such as Tris, glycine, etc. High concentrations of imidazole can also affect the reaction.
3) Labeling reactions are not recommended under strong light.
