FDG is a reagent used to detect beta galactosidase. Colorless and non-fluorescent FDG hydrolyzes to highly fluorescent fluorescein with excellent spectral characteristics (Ex/Em = 492/520 nm), matching the best detection window of most fluorescence instruments. Galactosidase-catalyzed hydrolysis of FDG results in increased fluorescence near 520nm. Alternatively, FDG can also be used to detect beta-galactosidase in a hair color pattern, since the enzyme product (fluorescein) exhibits a large extinction coefficient. FDG has been widely used in fluorescence microscopy and flow cytometry to identify LACZ-positive cells. It is also used to detect the expression of beta-galactosidase in living cells.
