Calcium measurement is essential for many biological studies. The jellyfish luminescent protein complex consists of 22kD meduse-carrying luminescent protein, molecular oxygen and luminescent coelenterin. When three Ca2+ ions bind to this complex, coelenterin is oxidized to coelenteramide, accompanied by the release of carbon dioxide and blue light. The luminescence intensity of cp, f or h medusa luminescence proteins containing coelenterin was significantly better than that of the deactivated medusa luminescence proteins reconstructed with natural coelenterin. Coelenterin h and cp have been used in HTS screening assay of GPCR.
