Mag-Fura-2 tetrapotassium salt is an intracellular magnesium indicator. This special magnesium indicator is also used to detect high concentrations of calcium.
Usage method (for reference only) : Preparation of storage solution 1~5mM storage solution is prepared by adding high-quality anhydrous DMSO, such as 50μg Mag-Fluo-4 AM(Mw:817.65 g/mol) dissolved in 12μL DMSO, after fully dissolved, 5mM storage solution is obtained.
【 Note 】 Unused storage solution is recommended to be stored at -20℃ in separate packages to avoid repeated freezing and thawing, and should be stored away from light. Use it as soon as possible.
Operation procedure
【 Note 】 The following methods are for reference only and should be adjusted according to the specific cell type and experimental requirements. The loading temperature range is 25 to 37 ° C (higher temperatures will increase the loading speed, but lower temperatures will minimize the internalization of the dye).
【 Note 】 The addition of surfactant Pluronic F-127 can promote the uniform dispersion of AM probe in water-soluble buffer.
1 Preincubate cells at 25℃~37℃ for 10min.
2 Take 4μL Mag-Fluo-4 AM storage solution and 5μL 20% Pluronic F-127, add 1mL cell culture solution, and mix vigorously to obtain a uniform probe dispersion solution.
3 Take 250μL of the probe dispersion prepared above and add it into 750μL of cell-containing medium and mix it well to obtain the working solution with a final concentration of 1~5μM.
4 The probe was incubated for 15 to 60 minutes.
5 The cells were cleaned 3 times, and then incubated for 30min to ensure complete de-esterification of the probe inside the cells. After that, the appropriate instrument is selected for fluorescence determination.
