Determination of Significance:
"Malate synthase (EC2.3.3.9) is a class of acyltransferases in transferases, mainly found in plants and microorganisms. It is one of the key enzymes in the glyoxylic acid cycle, which reacts with acetyl-CoA to form malic acid catalyzed by MS.
Measurement Principle:
MS catalyzes the production of malic acid by acetyl CoA and glyoxylic acid, along with the generation of coenzyme A, which converts colorless DTNB into yellow TNB with a characteristic absorption peak at 412 nm, from which MS activity can be calculated.

Visible spectrophotometer, 1 mL glass cuvette, balance, cryogenic centrifuge, water bath, mortar/homogenizer/cell sonicator, ice, and distilled water.
