OCT Compound is a water-soluble mixture of polyethylene glycol and polyvinyl alcohol and has been widely used in immunohistochemistry laboratories, it can support the tissue to increase its continuity and reduce wrinkles and fragmentation. Because OCT Compound is water-soluble, it will be soluble in water when bleach, so the background staining will not be increased in the later staining.Pre-infiltrating tissue by OCT Compound (opti-mum cutting temperature compound), then slice in cryostat .
Procedure:
1.take the tissue from sucrose solution, put it in mixture solution which 20g/L sucrose and OCT compound volume ratio is 1:1.
2. Transfer to OCT compound for 4h at room temperature, replace with new OCT compound and soak at room temperature for 6h.
3.Put the specimen on a tray, secice in a cryostat (generally is 22℃), thick is 22um, stick on pre-treated slides and store in-20℃ refrigerator.
