T4 DNA Ligase(Fast) is produced by Escherichia coli harboring the T4 phage gene 30. This enzyme catalyzes the formation of a phosphodiester bond between the 5' -phosphate group and the 3' -hydroxyl group between double-helical DNA or RNA. "This enzyme repairs single-strand breaks between double-stranded DNA, RNA, or DNA/RNA complexes, and can join DNA fragments with sticky or flat ends, but is inactive against single-stranded nucleic acids." It is mainly used for cloning DNA fragments of restriction enzyme products, site-directed mutagenesis and PCR product cloning, linear DNA self-looping and repairing double-stranded DNA defects. T4 DNALigase(Fast) requires ATP as a cofactor and takes only 10 min to complete the cohesive end joining reaction at room temperature.
Unit definition
The enzyme of 1 Weiss unit catalyzed the conversion of 1 nmol of [32 PPi] to the activated carbon adsorbed state in 20 min at 37 ° C. One Weiss unit is equivalent to approximately 200 sticky end joining reaction units (CEU), which is equivalent to ligation of a 50% HindiII-digested λDNA fragment in 30 min at 16 ° C.
