Collagen Fiber is the most widely distributed and abundant fiber in connective tissue, widely distributed in various organs, of which type I collagen fiber is mainly bone, skin and tendon; Type II collagen fibers are mainly cartilaginous collagen. Type III collagen fibers are mainly found in embryonic tissue, adult blood vessels and gastrointestinal tract. Type IV collagen fibers are mainly in the basal membrane. Sirius red and its interlining dye are strong acid dyes, easy to bind with the basic groups in collagen molecules, and adsorb firmly. By polarizing light microscopy, collagen fibers have positive uniaxial birefringent light properties. When combined with Sirius red composite dyeing solution, it can enhance birefringence and improve resolution, so as to distinguish the two types of collagen fibers.
Modified Sirius red staining solution (collagen fiber staining), G1472-reagent B, is mainly used for the study of abnormal changes of collagen fibers in various tissue lesions. The collagen fibers of heart vessels and other tissues are dyed red under the ordinary optical microscope, which is helpful for the classification and grading of various fibrotic lesions under the polarized light microscope.
