| Product Description | Strain LBA4404 has Ach5 background, and the nuclear gene contains a screening tag, Rifampicin resistance gene Rif. In order to facilitate the transformation operation, this strain carries the octocline Ti plasmid pAL4404, which has no self-transport function. This plasmid contains the vir gene (the vir gene is a necessary element for T-DNA insertion into the plant genome, and the T-DNA transfer function of the pAL4404 plasmid itself is damaged, but it can help the transferred binary vector T-DNA transfer smoothly). The pAL4404 Ti plasmid contains a screening tag: Strain LBA4404 was given streptomycin resistance by Strep, which was suitable for transgenic operation of tomato, tobacco and other plants. The conversion efficiency was up to 103CFU/μg by plant double pCAMBIA2301 plasmid detection, and the conversion efficiency did not change after 12 months of storage at -70℃. |
| Note | 1. When mixing the plasmid，it should be done gently，and the volume of the plasmid added should not be greater than 1/10 of the volume of the receptive state. The conversion efficiency will be sharply reduced if the plasmid is not pure or very large. 2. When there are too many colonies on the plate，the colonies are small. In order to obtain a large colony，the amount of plasmid used should be reduced or the amount of coating should be reduced，or the colony should be transferred to a new plate for growth. 3. The working concentration of rifampicin should not be higher than 25 μg/ml，too high rifampicin concentration will reduce the growth rate and conversion efficiency. 4. Rifampin has the effect of preventing the growth of miscellaneous bacteria and screening Agrobacterium;Adding streptomycin or gentamicin according to the resistance of the strain used can prevent the loss of Ti plasmid，but streptomycin is not conducive to the transgenic operation of Agrobacterium，so the addition of streptomycin or gentamicin can not be considered in the general culture of agrobacterium，and the probability of Ti plasmid loss is very low (negligible). |
