| Product Description | Agrobacterium LBA4404 has Ach5 background, and its nuclear gene contains rif, a screening gene for favorable fampicin resistance. In order to facilitate the transformation operation, this strain carries pAL4404, a Ti plasmid of octopine type without its own transport function, which contains the vir gene (the vir gene is the necessary element for T-DNA insertion into the plant genome, and the T-DNA transfer function of the pAL4404 plasmid is damaged. But it can help the transferred binary carrier T-DNA transfer smoothly). The Ti plasmid pAL4404 contained the screening tag gene strep, which conferred streptomycin resistance to LBA4404 strain. Some expression plasmids, such as pGreen, pGreenII-62SK and pGs2, which were missing replication-related elements (such as the REP region of pVS1 or the STA region of pVS1 plasmid), could not reproduce in LBA4404 strain. The helper plasmid pSoup can help these incomplete binary expression plasmids replicate in Agrobacterium Tumefaciens and render the LBA4404(pSoup) strain tetracycline (Tet) resistant. Suitable for transgenic manipulation of plants such as tomatoes and tobacco. Agrobacterium LBA4404(pSoup) receptive cells were prepared by special technology and detected by pGs2 plasmid. The conversion efficiency was up to 103CFU/μg, and the conversion efficiency did not change after 12 months of storage at -70℃. |
