The annealing extension temperature should be optimized according to the Tm value of the primer probe and the actual situation of the reaction. 4. The three-step method is recommended for primers with low annealing temperature or for amplification of long fragments over 200 bp. 5. Different target genes to be tested have different utilization efficiency of dUTP and sensitivity to UNG enzyme. Therefore, if the detection sensitivity decreases due to the use of UNG system, the reaction system should be adjusted and optimized. 6. Before and after amplification, please use special areas and pipettes, wear gloves to operate and change frequently; Do not open the reaction tube after the PCR reaction is completed to minimize the contamination of the PCR products to the sample. |
