| Note | (1) The pre-denaturation condition of PCR procedure was 95℃ for 5 min to fully lysate blood cells and denaturate nucleic acid templates. (2) It is recommended to use high-quality primers. (3) Increase the number of PCR cycles. (4) Reduce the annealing temperature, and try the annealing temperature gradient if necessary. Extending the annealing time to 90 sec~3 min can increase the amount of amplified products. (5) The amplification performance and specificity of primers were examined. (6) The amount of whole blood template used shall be adjusted within the range of the whole blood added by the above recommender. (7) When the product is too long, the primer needs to be redesigned, and the product length should not exceed 1.2 kb. (8) Extend the extension time within the cycle. |
