Indocyanine green (ICG) is a cyanine dye used in medical diagnosis. It is used to determine cardiac output,liver function,and hepatic blood flow,as well as for ophthalmic angiography. It has a peak spectral absorption close to 800 nm. These infrared rays penetrate the retinal layer,allowing ICG angiography to have a deeper circulation pattern than fluorescein angiography. ICG has a half-life of 150 to 180 seconds and is excreted from bile fluid only through the liver. Studies have shown that ICG targets atherosclerosis within 20 minutes of injection and provides sufficient signal enhancement for the detection of lipid,inflamed,coronary artery-sized plaques in atherosclerotic rabbits. Ex vivo fluorescence reflectance imaging showed that atherosclerotic rabbits injected with ICG had a higher plaque target-to-background ratio compared to atherosclerotic rabbits injected with normal saline. Amino-reactive ICG derivatives are used to prepare ICG bioconjugates with antibodies and other biomolecules. All ICG dyes need to be dissolved in DMSO.
