Glutathione peroxidase 4 (GPX4, EC 1.11.1.12) is one of the peroxidases with selenocysteine as the active center. It is a member of the glutathione peroxidase family. It can catalyze glutathione (GSH) to react with peroxides and reduce peroxides to hydroxyl compounds, thereby protecting cells from free radical damage.
Glutathione peroxidase catalyzes the reaction of GSH with peroxide to form oxidized glutathione (GSSG). GSSG reacts with NADPH to form GSH under the action of glutathione reductase (GR). NADPH has a characteristic absorption peak at 340 nm. The rate of decrease in absorbance of the reaction system is linearly related to the activity of glutathione peroxidase. GPX4 inhibitor was added to the reaction system, and GPX4 specific enzyme activity is calculated by measuring non-specific enzyme activity and total enzyme activity.
