Product introduction
Uracil DNA glycosidase (UNG, UDG) is a 25kDa enzyme that catalyzes the release of free uracil from single-stranded and double-stranded DNA containing uracil.
Scope of application
The UNG enzyme has no activity against RNA and is mainly used to prevent contamination of PCR amplified products.
Unit definition
The amount of enzyme used to degrade 1μg of single-stranded DNA containing dU bases for 1 hour at 37 ° C was 1 unit.
Optimum pH
8
Optimum temperature
25℃
Matters needing attention
1, avoid multiple freezing and thawing, do not be exposed to large temperature fluctuations. Temperature fluctuation has a great influence on the stability of the product.
2. UNG enzyme can remove accidentally contaminated PCR products containing dUTP before PCR reaction, thus avoiding false positive results caused by contamination;
3. Because different genes to be amplified have different utilization efficiency of dUTP and sensitivity to UNG enzyme, the reaction system should be adjusted and optimized if the detection sensitivity is decreased due to the use of UNG system.
4. Use special areas and pipettes before and after amplification, wear gloves to operate and change frequently, and do not open the reaction tube after the PCR reaction is completed. To minimize the contamination of samples by PCR products.
