McManus first used the periodate Schiff technique to display mucin in 1946.It is often used to display glycogen and other polysaccharides, neutral mucous substances and some acidic substances, cartilage, pituitary, fungi, pigments, amyloid substances, basement membrane and so on.Periiodic acid (also known as periodate) is a strong oxidant that can oxidize 1,2-ethanediol groups in sugars and related substances, turning them into dialdehydes. Aldehydes can combine with Schiff reagent to form a fuchsin compound, producing a purple red color. Due to the fact that periodate can also oxidize other substances within cells, pay attention to select the concentration and oxidation time of periodate when using it, so that oxidation can be controlled within the range of oxidizing ethylene glycol groups into aldehyde groups without peroxidation, which is a crucial step.
