Percoll separation solution is a kind of silica gel particles treated with polyvinylpyrrolidone, which can form a continuous density gradient after high-speed centrifugation. Because of the low diffusion constant of Percoll, the gradient is very stable. When the pre-formed density gradient was used in Percoll separation solution, satisfactory cell separation results could be achieved at low centrifugal force (200~ 1000 g) in ten minutes. In addition, Percoll does not penetrate the biofilm and is not toxic to cells, so it is widely used to isolate cells, subcellular components, bacteria and viruses, and to separate damaged cells and their fragments from intact living cells. Percoll is often used to isolate and communicate plant protoplasts, nuclei, chloroplasts, and mitochondria.

Percoll is an ideal medium at present because of its low permeability, non-penetration, high density and non-toxicity. Percoll density gradient centrifugation has been successfully used to separate animal cells and their organelles for many times. More than ten kinds of animal cells including human blood cells, bone marrow cells, red blood cells, white blood cells, lymphocytes, hepatocytes and so on have been purified.
