The single isomers of ROX are good choices for labeling peptides and nucleotides，as they provide better resolution in HPLC purification，which is often required in bioconjugation. The variable ratios of the 5- and 6-isomers often complicate the interpretation of labeling results and assay performance. 5-ROX is mainly used to label peptides and proteins，and compared to other rhodamines，ROX is very unstable and care must be taken to prevent degradation during storage.
