Tetramethylrhodamine-5(6)-maleimide is a very good fluorescently labeled probe that modifies peptides，nucleotides，nucleic acids，and proteins by thiols to form bioconjugates with orange fluorescence. Maleimide is a commonly used reagent for thiol modification. In most proteins，the site of the thiol reaction is at a cysteine residue，which is intrinsic to the protein and results in a decrease in cysteine in the protein. Unlike iodoacetamide，maleimide does not react with histidine and methionine under physiological conditions.
