Preparation method :
1. Weigh 2g of salmon sperm DNA in a 500ml beaker and add about 400ml of TE Buffer.
2. After dissolving, 8ml of 5 M NaCl was added to a final concentration of 0.1 M by stirring with a magnetic stirrator for 2 to 4 hours at room temperature.
3. DNA was cut by rapid aspiration of the above solution about 20 times using a 17-gauge hypodermic needle.
4. Extract once with phenol/chloroform and once with chloroform.
5. Ethanol precipitation was performed by adding 2 times the volume of precooled ethanol.
6. The DNA was recovered by centrifugation and dissolved in 100 ml of deionized water, and the OD260 value of the solution was determined.
7. After calculating the DNA concentration of the solution, the DNA solution was diluted to 10 mg/ml.
8. After boiling for 10 minutes, divide into small portions (1 ml/ portion). The samples were stored at -20 ° C.
9. After heating in a boiling water bath for 5 minutes before use, cool quickly in an ice bath.
