5-fluorouracil, referred to as fluorouracil, is a pyrimidine antimetabolite and an antineoplastic drug. This product is only used for biochemical research.
5-fluorouracil Principle:
5-fluorouracil antineoplastic drugs are a class of broad-spectrum antineoplastic drugs that interfere with the nucleic acid metabolism of tumor cells through a variety of pathways and metabolites. The main pathways are: 5-fluorouracil is converted into 5-fluorouracil deoxynucleotide (5 F-dU old) in tumor cells, which can covalently combine with reduced tetrahydrofolate and thymidine nucleotide synthetase (TS) to form a ternary complex, which inactivates TS acid and inhibits DNA synthesis, thereby inhibiting tumor cell proliferation.
The drug is mainly specific for s phase, but after being converted to 5-fluorouracil nucleoside (5-FUR) in vivo, it can also be incorporated into RNA to interfere with protein synthesis, so it can also act on cells in other phases. As an anticancer drug, 5-fluorouracil has an inhibitory effect on a variety of tumors, but the disadvantage is that the effective dose of 5-fluorouracil is similar to the toxic dose, which kills cancer cells while causing serious damage to normal cells.
