G418 is an aminoglycoside antibiotic, which is similar in structure to neomycin, gentamicin and kanamycin, and is the most commonly used resistance screening reagent for stable transfection in molecular genetic tests. It blocks protein synthesis by inhibiting the gene for the transposon Tn601, Tn5, interfering with ribosome function and producing toxins to prokaryotic and eukaryotic cells, including bacteria, yeast, plant and mammalian cells, but also protozoa and worms. When neo gene is integrated into the DNA of eukaryotic cells, the sequence encoded by neo gene can be transcribed into mRNA, thus obtaining the efficient expression of the resistance product aminoglycoside phosphotransferase, making the cells resistant and able to grow in selective media containing G418
Description:
Antibiotics for eukaryotic expression screening can be used to screen stable cell lines expressing specific resistance genes. For screening mammalian cells, the recommended initial screening concentration is 400 μg/ml. The recommended initial screening concentration for plant cells is 10 μg/ml. Screening yeast, recommended starting concentration of 500 μg/ml. According to the effect of the initial concentration, the concentration of G-418 can be appropriately increased or decreased.
Use and synthesis method:
Blocking peptide synthesis by interfering with ribosome function. It is not a commonly used standard antibiotic and is often used to select prokaryotic and eukaryotic cells transfected with neomycin resistance genes. It is widely used in gene transfer, gene knockout, resistance screening and transgenic animals.
Feature:
Soluble in water, methanol, buffer and medium. After dissolution, 0.22μm filtration to remove bacteria, divided into small packages, 4℃ can be stored for 12 months, longer storage time at -20℃. The most commonly used storage solution is formulated with 100mMHEPES(pH7.3) at a concentration of 50mg/ml. The advantage of using HEPES formulation is that the addition of storage solution does not change the pH of the culture system.
