saline sodium citrate (SSC) buffer solution is the most standard imprinting and hybridization treatment solution in molecular biology, designed for various hybridization experiments to achieve denaturation and cleaning purposes. The main components are sodium chloride and sodium citrate. Sodium citrate in SSC buffer acts as a buffer, and salt ions (Na) neutralize the negative charge on the nucleic acid main chain to make it neutral, so that the binding of probe and target sequence is easier. Also used for SDS-PAGE electrophoresis separation gel preparation. For nucleic acid hybridization, different concentrations have different effects:
Commonly used 2×, and 0.5×2×SSC: high-salt washing membranes to wash away some of the non-specific binding probes.
0.5×SSC: Low salt washing membrane, increase the tightness of nucleic acid chain, so that the repulsion between RNA/DNA increases.
This product is 20× concentrated liquid, which can be diluted directly with deionized water according to experimental requirements.
Note:
1. Pay attention to avoid microbial contamination of the solution during use.
2. Store at room temperature, if the solution is kept at a low temperature, it may precipitate and can be shaken or heated to dissolve.
